Part of Project Purification's goal was to maintain the "Divinity of Mankind," which called for the eradication of all non-vital human prosthetics and mutations. Stukov was part of the fleet sent by the United Earth Directorate to subjugate the sector and the powers there, terran and alien. However, Stukov, Kerrigan and many terrans feel that the UED is still out there watching and waiting, and that a second invasion is on the horizon.

Both Zarya and Stukov are from Earth and Russian. Stukov's use of old Russian phrases, and the UED's use of classic relics from the 20th century such as record players, shows a nostalgia for the past even in 2500.

Stukov has constantly been tied to the zerg since his invasion of the sector, first setting the stage for his fleet to control the Second Overmind, then dying to the agent of Kerrigan, Samir Duran, then being infested by the cerebrate Kaloth, then briefly cured, and as the cure failed he was brought back into the Swarm under Kerrigan.

While part of the Swarm, Stukov has retained his own individuality and is disconnected from the hive mind, likely due to the brief curing of his infestation in StarCraft 64.

In StarCraft: Brood War, Alexei Stukov's faction of the UED appears in-game using the standard terran units (though with the two additional units added in the expansion). However on maps where both the Terran Dominion and UED are present, the UED's AI will strongly favor building air units and infantry over factory units like siege tanks.

Many infested are in perpetual pain. In his Heroes of the Storm interpretation, Stukov can grow his arm for his heroic and Lurking Arm abilities.
On, the plus side, I've gotten very good at arm wrestling lately. A reference to how large only one arm of Stukov has gotten.

While Duran was experimenting on me, I used to tell myself it could always get worse. Then, it got worse. Duran was a xel'naga who first killed Stukov, then when he was resurrected by infestation imprisoned him under the guise of researching the protoss cure. He used Stukov in order to help accelerate his research on creating protoss/zerg hybrids, torturing him for years.
A virus is neither alive, nor dead. It is... other. Existing only to plague and control the living. <beat> I know the feeling. Stukov has been resurrected via infestation after complete death, and spreads the zerg hyper-evolutionary virus around to others to both infest, and in Heroes of the Storm, to heal.

Kaneda? Who is that? And why do I want to crush him? A reference to the character Tetsuo from the manga and movie Akira. Much of Stukov's kit, including the horrific extending and mutating arm, is based off of Tetsuo. Kaneda is Tetsuo's old friend, who quickly becomes his enemy in the story.
For my enemy, the day Stukov infested their team was the most horrifying experience of their lives. To me, it was Tuesday. A reference to a line from the character M. Bison from the Street Fighter movie, "for you, the day Bison graced your village was the most important day of your life. But to me, it was Tuesday." Bison and Stukov have similar uniforms and hat wear.

However, due to the machinations of Kerrigan and Samir Duran, DuGalle was tricked into ordering Duran to kill his old friend. After his fleet's defeat and imminent destruction, DuGalle wrote a message to his wife expressing how deeply his guilt ran for killing Stukov, and committed suicide. After his resurrection and infestation, Stukov regularly states how much he misses his old friend.
